Abstract:
One tube osmotic fragility test (OFT) in 0.36% normal saline solution is widely used in thalassemia trait screening. Here, we studied the 2 minutes osmotic fragility (2 MIN OF) value in 0.45% glycerine saline solution of various severe thalassemia traits, 50 cases each. The erythrocyte lysis was recorded photometrically at 620 nm, 15 and 120 seconds after mixing erythrocyte with glycerine saline solution. The result showed that 60% 2 MIN OF could be used for differential screening all of beta-thalassemia trait and alpha-thalassemia-1 trait (mean +/- S.D = 30.5 +/- 1.4% and 30.8 +/- 1.2%, respectively), but only 60% of Hb E were detected. However, combination with the simple Hb E microcolumn chromatography technique all severe thalassemia traits could be effectively detected.
